Yeah I found out I didn't need the puller and instead needed a holder.  I actually got around the holder by using an impact gun.  The nut came off so easily that way.  As for the sprockets I have not tried them yet but I plan on picking up a 51 tooth for the rear.  I have noticed the flywheel weight helping because I went riding yesterday, but I didn't do anything too technical yet so I don't know how much of a difference it has made yet.

I have a 2005 YZ250F.  At first I put a 12/49 on it.  It was a ton better.  then i added a 10oz flywheel weight.  This greatly improved its trail abilities.  I found a good used Rekluse Z start local for cheap and decided to try it.  It makes my bike unbelievable on the trails.  My son has a 2004 without any mods and I sit and watch him stall it and spin the tire while I can crawl.  any of the changes will help,  but for me all the changes made mine exactly what i was looking for.  Also when you are riding along you dont even notice the rekluse or the flywheel weight.  It still pulls the front wheel up effortlessly and you dont loose power.  It just moves the power to a different RPM

Next time you are doing an upper end...split the cases and put in the WR 1st gear.  I did this on my 06.  You never use 1st on a MX track...so who cares what it is.  Putting in the WR 1st gives you a MUCH lower 1st gear....
