I just got a 2001 YZ 426, it starts first kick everytime when its cold and runs great. Is it worth checking the valves on it. Is it a big job to check the valves? Not sure how many hours are on it. Just wondering if its worth doing if it starts and runs well.

Better safe then sorry. Checking valves is not a hard thing to do.

What possible harm (ignoring stripped screws and the like) can come from checking valves, changing oil, looking for loose spokes, inspecting the forks seals, etc.?

This stuff is known as routine preventative maintenance for a reason.
