Stroker recommends the following.
What are the hetting specs for the 2000 WR 400?
StockJetting StrokerRecommends
42 Pilot 50
DRS Needle DVR

3rd Clip Position 4th
165 Main 180
75 Pilot Air 100
200 Main Air 200
1 ½ out Fuel Screw 2 out
1. NOTE: Leave air box lid off! These specs are with Stroker exhaust or stock WR muffler with noise baffle out. We recommend using Stroker dual sport quiet tip rather than noise baffle out.

Has anyone tried this combo. How does this compare to Taffy jetting?

I can only tell you what I have experienced with Stroker. The settings they give you will put you in the ballpark. Every bike is a bit different and yours may require some fine tuning from their jetting specs.

Is your bike still WR timed??
Do you have an aftermarket pipe on it??
