stock 'R' carburetor question

the tube (?) that the main jet seats in, I guess you could call it the needle guide maybe, has a nut on it to adjust its hite into the bore of the carb. What exactly does this effect and does it need to be kept at the stock position?

I am finally getting around to changing to the B53E needle and swapping out the brass piece that came with it, just want to make sure I'm not going ot put it together wrong and have to strip the whole thing back down again.

Snug the carrier tube all the way down and then tighten the jam nut...
